nCounter RLFs, Probe Annotation Files, and Gene Lists

Reporter Library File (RLF): Used by the nCounter system as a “lookup table” to convert the molecular barcodes to the assigned genes. RLFs for our off-the-shelf (not custom) panels are listed below.

To run Panel Plus or CodeSet Plus assays, the Panel or CodeSet RLF must be merged with the Panel Plus or CodeSet Plus ALF (Add-on Library File). To obtain a merged RLF file, email Bioinformatics.BSB@Bruker.com. Include the original RLF and ALF. Failure to use a merged RLF will result in no count information being collected for targets of Plus products.

WARNING: Using the wrong RLF can result in scrambled data. Please double check that the correct RLF and panel pairing are being used. Be especially mindful of Human vs Mouse.  Check after the run that the data looks as expected.  
If the wrong RLF is used: 

  • On the SPRINT Profiler – The data can be re-mapped to the correct RLF using the Fix RLF feature.  See the SPRINT User Manual for details.
  • On the Pro/MAX/FLEX – The cartridge must be re-scanned with the correct RLF. 

Please note that GeoMx DSP assays with nCounter readout use a GeoMx-specific RLF that is installed on the nCounter Analysis System during GeoMx DSP training.

Probe Annotation File: Download and use in setting up the nCounter Data Analysis Pipeline. With the launch of the nCounter Analysis Pipeline, file formats have changed. Probe Annotation Files used with nSolver are not compatible with the new Pipeline. Please download the correct file for your panel below and use it when setting up the nCounter Analysis Pipeline.
