Characterization of Key Tissue Structural Features

Providing a modular building block for essential spatial discovery of key structural features of human FFPE samples, the Tissue Architecture Assay Kit includes a panel of antibodies curated and optimized by our robust validation workflow. The panel targets key epithelial, lymphatic, lamina, vasculature and connective tissue biomarkers and can be expanded by pairing with additional VistaPlex Multiplexing Assay Kits or your own markers of choice.

The VistaPlex Tissue Architecture Assay Kit enables automated staining and imaging of 10 human FFPE samples for 12 biomarkers using the CellScape™ Precise Spatial Proteomics platform. An online protocol is available to guide consistent and reliable execution of the multiplexed assay.

How it Works

The VistaPlex Tissue Architecture Assay Kit enables multiplexed detection of key tissue architecture markers directly in situ, delivering quantitative, spatially resolved phenotypes across FFPE tissue. Each panel is fully validated for use with EpicIF™ chemistry and the CellScape™ platform, ensuring consistent staining, robust signal removal, and high-quality single-cell proteomic data.

Validated Antibodies

The kit includes 12 validated, fluorescently labeled antibodies to support identification of structural tissue features.

Optimized Protocol

An online protocol is available to guide consistent and reliable execution of the multiplexed assay.

Reliable Performance

Antibodies for the multiplex panel were validated using eight human tonsil FFPE tissue samples to ensure specificity, sensitivity, and reproducibility. Additional FFPE tissue types were validated for broad sample suitability.

Expandable Assays

Expand the Tissue Architecture Assay Kit with modular VistaPlex Assay Kits to build a customized Precise Spatial Proteomics panel. Compatible kits can be run on the same sample, and the CellScape workflow lets you seal, store, and revisit the same slide. Add new biomarkers later to drive data-driven panel expansion and make the most of precious samples.

Intra- and Inter-Site Validation

Following clone selection and assay design, the multiplex panels are validated for performance by specificity, sensitivity, reproducibility, and suitability measurements. Validation experiments are completed using serial tissue sections from multiple relevant tissues, on different CellScape instruments, in different laboratory sites, and with staining scored by multiple scientists.
Qualitative Suitability and Specificity Assessment
Average scores from technical replicates of human FFPE Tonsil are visualized in a heatmap (A, green=pass, red=fail) and a radar plot (B). n = 8 samples scored by four independent judges.
Quantitative Reproducibility Assessment
Cell density measurements for each stain across three technical replicates of human FFPE tonsil.
Tissue Type Suitability

Validation is performed on human FFPE tonsil as a positive control, as well a multiple fundamental disease states.
